由於在細胞培養過程中,細胞的生長與代謝會使培養液的環境產生劇烈的pH值變化。為了維持讓細胞穩定生長的環境,就需於培養液中添加適量的生物緩衝劑。然而,市面上能用於細胞培養的生物緩衝劑以及替代方式多不勝數,為什麼要選擇HEPES呢? 一般來說,大多數的細胞所適宜之培養環境為pH7.2-pH7.4[1][2],而HEPES緩衝劑應用範圍正好落在pH6.8-pH8.2,符合利於細胞培養時所需之pH值特點。且相較於PBS、TRIS等其他緩衝劑,在維持細胞培養液的pH值環境中,HEPES具有較高的穩定性[3][4][5],也因此HEPES緩衝劑廣泛應用於細胞培養、組織培養、蛋白質純化與提取、免疫沉澱、細胞裂解、活細胞成像等生物與生物化學研究中。
