Hopax is a manufacturer and supplier of TRIS, also known as Trometamol (CAS 77-86-1), a zwitterionic biological buffer often used as a buffering agent in biological and biochemical research.


Uses of TRIS Buffer:

  • In buffer solutions for gel electrophoresis (TAE and TBE)
  • In the preparation of Laemmli buffer, one of the most popular running buffers for SDS-PAGE
  • As a binding buffer and eluent in anion exchange chromatography
  • For the evaluation of bacterial endotoxins
  • In capillary electrochromatography (CEC), since it has low ionic mobility
  • TRIS is commonly used as a buffer for vaccines, such as mRNA vaccines.
  • In personal care and cosmetics applications, TRIS functions as a buffer and a mild neutralizing amine for skin and sun care formulations. This chemical is considered an excellent emulsifying agent and viscosity controller, and it is known for its capacity to maintain the original colors of lotions after opening


Metal binding: (Compare with other buffers)

Strong interaction with:

Cr, Fe, Co, Ni and Cu
Weak interaction with:

Mg, Ca, Zn, Cd and Pb


Concerns:

  • It is not a very efficient buffer below pH 7.5.
  • It has a potentially reactive primary amine, which often acts as an inhibitor.
  • It has a tendency to permeate membranes, therefore it is not suitable for most cell culture work.
  • It is toxic for many mammalian cells.
  • It is not suitable for use with the Bicinchoninic acid (BCA) assay.
  • It may interfere with the Bradford dye-binding method of protein assay.
  • The pH of solutions containing Tris is very dependent on temperature, therefore it is important to prepare TRIS buffer at the temperature at which it will be used. (Read more)
